Tons of different categories that work differently but all require amazing movement, making for an entertaining playing and viewing experience.
For all 120 stars or just to complete the game and/or just to complete the story, although I consider that I guess that all 120 stars could be just to complete the game as well! Yeah, I can't do this because of Mario Wing over the Rainbow and other hard levels that will probably stop me from getting all of the stars and/or reaching Bowser and /or reaching the final boss Bowser! - HeavyDonkeyKong

Good game, only metroid game on the list so that's where my vote goes to.
MUCH better than Zelda in my opinion
Pretty obvious to be honest. More routes through the game than you can count, and one of the games directly responsible for the rise of speedrunning.

The Binding of Isaac: Rebirth ('Isaac') is a fairly hard game that for the average play often takes about 75 minutes to beat, and sometimes the pro players take that long. So speed runs of 10-20 minutes or less are pretty cool.
It's vague by 'Best' refers to here, but I sincerely think Isaac is the best game to speed run in terms of 1. How quickly it can be speed run, 2. How enjoyable it is to speed run the game, and 3. It's the best in making it Possible to speed run it, in giving the player the tools to do a speed run.
Isaac is a fast-paced game, and when you go even faster, its usually because you're more powerful and have awesome weapons to kill all your enemies. The games power-ups are random and varied. So if you get lucky, then the game could give you the most powerful items, which is what allows players to win in 4 minutes.

Due to a recent discovery (recent relative to 10/20/19), the game is now being pushed to its limits. The times will only continue to decrease, and the skill cap is incredibly high. Not only that, but ABH'ing (Accelerated Backwards Hopping) around is nigh ecstasy.

- Factory: 3-1 to 3-17
OR 3-1 to 3-11, then 3-14 to 3-19 (fastest route, do it only if you can do the 3-18 skip consistently)
- Hell: 4-1, then 4-3 to 4-17, then 4-19
- Rapture: 5-1 to 5-7, then 5-9 to 5-18 (fastest route, do it only if you can get the 5-1 fast strat and do the 5-18 consistently)
OR 5-2 to 5-18 (if you can't do 5-1 without dying/messing up)
OR 5-1 to 5-17 (old route, do it if you can't do 5-18)

The above routes assume standard PC loading times. If your load times are worse, most likely due to playing on console, you have some route options. In general, load times matter much more when you enter and exit stages from level select rather than simply beating levels and advancing to the next one, so routes that involve exits to map will become comparatively worse. How can you tell if your load times are worse? If you watch streams and see that nearly everyone else saves seconds over you every time you exit to map.
Super Meat Boy Any Wrestling
With bad loads, try these routes instead:
forest: 1 through 17
factory: 1 through 17 (borderline case; would be worth timing yourself, but 1-17 is easier)
hell: 1 through 17 (if you're willing to skip ahead of the moving platform before the end of 4-2, otherwise do normal route)
rapture: 1 through 17 or 2 through 18 (whichever is easier; compare your 5-1 time vs 5-18)
